摘要
为给裸燕麦β-葡聚糖的开发应用提供科学依据,以山西产裸燕麦为原料,研究了溶剂、温度、提取时间和β-葡聚糖酶水解等提取条件对燕麦β-葡聚糖分子量分布的影响。结果表明,较强的酸性和碱性条件会导致燕麦β-葡聚糖分子量下降,水提取β-葡聚糖的分子量较高。β-葡聚糖的分子量随着提取温度的提高而增加,随着提取时间的延长而下降。不同条件下提取的燕麦β-葡聚糖分子量分布在340-2 000 kD之间。β-葡聚糖酶水解会使β-葡聚糖分子变小,在几乎完全水解时分子量为2-5 kD。
In order to provide scientific basis for the naked oat uses, the naked oat produced in Shanxi were used as material, we used the method of Sepharose CL-4B gel-filtration column chromatography to determine the molecular weight(Mw) distribution of β-glucan extraction in different extracting conditions. The result showed extraction conditions could affect of the molecular weight of the β-glucan. Mw of β-glucan was significantly decreased by NaOH and HCI-KCI buffer(pHI. 5) extraction. Hot water extraction gave higher Mw of β-glucan. Increasing the temperature of extraction led to an increase in the Mw of β-glucan, but prolonging the time of extraction led to a decrease in the Mw of β- glucan. The distributions of Mw of β-glucan were 340~2 000 kD in different extracting conditions. β- Glucan(Mw〉2 000 kD) hydrolyzed by β-glucanase could lead to the decrease of Mw(2~5 kD).
