苯丙氨酸对慢性缺氧性肺动脉高压大鼠的保护作用 被引量:1

Protective effect of phenylalanine on rats with chronic hypoxia-induced pulmonary hypertension

摘要 目的:探讨苯丙氨酸(PHE)对慢性缺氧所致的肺动脉高压的影响及其可能机制。方法:28只雄性Wistar大鼠随机分为对照组(n=9)、缺氧组(n=10)、缺氧+PHE组(n=9)。缺氧组与缺氧+PHE组每d给予缺氧处理。缺氧+PHE组每d于缺氧前腹腔注射PHE400mg/(kg.d),对照组与缺氧组腹腔注射等量生理盐水,共21d。采用右心导管法测定右心室平均压(pMRV)及肺动脉平均压(pMPA),同时用多导生理记录仪测定心率和血压。测定右心室(RV)质量/(左心室(LV)质量+室间隔(S)质量)(mRV/(mLV+mS))比值,并观测肺血管形态结构的变化和各组血清血管内皮生长因子(VEGF)的变化。结果:缺氧组的pMRV、pMPA、(mRV/(mLV+mS)明显高于对照组(P<0.01)与缺氧+PHE组(P<0.05);缺氧组的肺细小动脉管壁厚度、管壁横截面积、管壁厚度占血管外径的百分比及管壁横截面积占血管横截面积的百分比均明显高于对照组与缺氧+PHE组(P<0.01)。缺氧+PHE组大鼠血清VEGF水平明显低于缺氧组(P<0.01),但仍高于对照组(P<0.05)。结论:PHE可能是通过降低VEGF的表达而降低慢性缺氧大鼠的右心室压及肺动脉高压,逆转右心室肥厚及血管结构的重建。 Aim: To explore the effect and mechanism of phenylalanine (PHE) on chronic hypoxia-indueed pulmonary hypertension in rats. MethOdS:Twenty-eight male Wistar rats were randomly allocated into control group (n = 9), hypoxia group (n = 10), and hypoxia plus PHE group (n =9). PHE was injected into abdominal cavity in the rats of hypoxia plus PHE group (400 mg/( kg · d)) before hypoxia for 21 d. Normal saline was given in both control group and hypoxia group equally. Right ventricular mean pressure (PMRV) and pulmonary artery mean pressure (PMPA) were evaluated using right cardiac catheterization. Heart rate and blood pressure were evaluated using multipurpose polygraph. The ratio of right ventricular mass to left ventricular plus septal mass (RV/( LV + S)) and the levels of VEGF in serum were determined. Meanwhile, microstructure of pulmonary arteries was observed. Results: PMPA, PMRV and RV/( LV + S) were significantly increased in hypoxia group in comparison with control group (P 〈 0.01 ) and hypoxia plus PHE group ( P 〈 0.05). The tubal wall thickness, and tubal wall cross section area of small pulmonary artery, the ratio of tubal wall thickness to vascular diameter and the ratio" of tubal wall cross section area to total vascular cross section area in hypoxia rats were significantly higher in comparison with those in control group and hypoxia plus PHE group (P 〈 0. 01 ). The levels of serum VEGF in hypoxia plus PHE group were significantly lower than that in hypoxia group (P 〈 0. 01 ), but still higher than that in control group (P 〈 0. 05). Conclusion : PHE can effectively reduce the level of PMPA and PMRV of chronic hypoxia rats, and ameliorate pulmonary vascular structural remodeling and right ventricular hypertrophy, which may relate to its reducing the level of VEGF in serum.
