僵蚕提取液中蛋白质和草酸铵等成分的定量分析 被引量:28

Quantitative Analysis of Protein and Ammonium Oxalate in the Extract Liquide from Bombyx Batryticatus

摘要 目的对僵蚕提取液中三种形态的含氮物质、草酸盐、总离子浓度和还原性物质进行含量测定。方法采用离子交换和经典化学分析方法。结果僵蚕提取液中固形物含量为38.52%,RSD=0.92%(n=3);总氮、铵态氮和酰胺态氮含量分别为5.798%、0.868%、0.196%,蛋白质含量为29.74%,占总固形物的77.2%;草酸铵含量为3.85%(不包括0.96%的草酸根对应的其他盐),占总固形物的9.98%;总离子浓度按一价离子计为1.099mol/L;能被高锰酸钾在酸性条件下氧化的还原性物质含量相当于1.015mol/L的草酸盐浓度,其中阳离子型、阴离子型和非离子型还原物质浓度占总还原物质的摩尔百分率分别为7.84%、64.16%和不低于28.0%。结论本研究为进一步研究僵蚕提取液的抗凝作用和药理作用机理奠定了一定的物质基础。 Objective To determine three nitrogenous compounds, oxalate, ions concentration and reducive substance in the anti-coagulant extracting solution from Bombyx Batryticatus. Methods The chemical analysis and ion exchang methods were used to the determinations. Result The content of dry extract was 38.52% with RSD=0.92% (n=3). Total nitrogen, ammonia-state nitrogen and acylammonia nitrogen was 5.798%, 0.868% and 0.196%, respectively, and amounted to 29.74% proteins that was 77.2% of dry extract. In additions, the content of ammonium oxalate was 3.85%, and amounted for 9.98% of dry extract. The total ion concentration by univalent ion was 1.099 mol/mL. The reducing substances that can be oxidized by KMnO4 equal to the 1.015 mol/mL by oxalate, and positive ion, negative ion and neutral molecules in these were 7.84%, 64.16% and over 28.0%, respectively. Conclution This work will benefit more studies of anti-coagulant extract solution from Bombyx Batryticatus.
