摘要
目的:建立兔舌癌模型并观察颈淋巴结及其肺转移。方法:采用J-IN2肿瘤细胞株接种的方法。结果:2周后成功的建立了兔舌癌模型。成癌率100%(10/10),动物模型经历了肿瘤迅速增长、肿瘤中心坏死、表面破溃等阶段。舌癌晚期,舌癌模型出现恶病质状态,其中8只兔出现颈淋巴结转移(转移率80%),4只出现了肺转移(转移率40%),平均生存期72.5d,组织学表现为低度分化的鳞状细胞癌。结论:通过J-IN2肿瘤细胞株接种而建立的兔舌癌模型具有在短期内易建立、稳定、重复性好、成癌率高、生存期长等特点,晚期发生颈淋巴结转移和肺转移,临床表现同人舌癌相似,为舌癌的生物学、治疗学研究,提供了一个大型的良好动物模型。
Objective: To establish the rabbit model of tongue carcinoma and observe its metastasis to cervical lymphonode and lung. Methods: Tumor cell strain J - IN2 was inoculated. Results: The model of tongue carcinoma in rabbits was established successfully after 2 weeks. The rate of carcinogenesis was 100 % (10/10). The animals experienced stages including tumor rapid growth, necrosis in center and ulcer in surface. 8 of 10 rabbits occured metastasis, the rate of metastasis was 80 %, and 4 of 10 occured pulmonary metastasis. The average life span was 72.5 days. The histological type of carcinoma was squamous cell carcinoma. Conclusion: The inoculation of tumor cell strain J - IN2 is a feasible and repeated method on establishing the model of tongue carcinoma, with a high rate of carcinogenesis and long lifespan. The advanced carcinoma metastases to cervical lymphnode and lung. The signs of animal model are similar to those of human, which provides a satisfactory animal model for study on biology and therapeutics in tongue carcinoma.
