摘要
目的探讨心肌梗死后心肌细胞核分裂指数、端粒长度的变化及其生物学意义。方法结扎大鼠左冠状动脉制备心肌梗死模型,观察大鼠心肌梗死后血流动力学改变;HE染色评估梗死面积;荧光免疫法检测核分裂指数;Southern杂交测定端粒片段平均长度。结果大鼠心肌梗死后第14天及第30天端粒酶表达明显增加(P<0.05);端粒平均长度在心肌梗死后第14天较假手术组及心肌梗死后1d分别增加了1.08倍(P<0.05)。结论大鼠心肌梗死后心肌细胞端粒酶表达增强,端粒长度改变,二者可能参与了心脏的生长储备。
Objective To study cardiomyocyte mitotic index and the telomere length after myocardial infarction. Methods Rats' Myocardial infarction ( MI ) models in rats mimicked by ligation of the left coronary artery. Hematoxylin-eosin stain was used to assess the infarct size. Immunofluorescence microscopy was used to examine the change of the myocyte mitotic index. Southern blot analysis was used to observe the average length of the telomere fragments. Results At 14th and 30th day,the myocyte mitotic index were significantly increased( P 〈 0.05). The average length of telomcre fragments was 1.08-fold in heart of 14th day compared with the sham group at 1st day after MI,respectively ( P 〈 0.05 ). Conclusion The increased expression of telomerc and the changes of telomere length may affect the heart's growth capacity.
