摘要
采用传代稳定,高产功能性多糖的桑黄菌株,对其进行产糖营养条件优化。首先采用PDA培养基进行菌种活化,然后通过单因子实验最终确定出最为适宜的培养基成分和发酵条件,并且采用正交试验设计进一步优化培养基配方。结果表明,可溶性淀粉是最佳碳源,豆粕是最佳氮源,桑黄真菌优化培养基配方为5%可溶性淀粉、2%豆粕、0.1%磷酸二氢钾、0.05%硫酸镁、氯化钙0.05%,鸟苷酸0.03%、植物油1%。最适菌丝体生长的液体发酵条件为:培养温度为28℃,摇瓶转速160r/min,pH值6.5,装液量(250mL三角瓶)为70mL,发酵时间为7d最佳。菌丝干质量从原来的1.00g/100mL优化到1.76g/100mL,多糖含量提高了53.68%。
Phellinus igniarius (L.EXfr.)Quel is a medicinal fungus producing intracellular polysaccharide. In this article we used phellinus igniarius with a stable generation characterized. We selected and optimized the best nutrition conditions and fermented conditions for this fungus. First, we inoculumed the phellinus igniarius on the PDA culture medium in order to active the fungus. Second, we selected the best components of the culture medium and the ferment conditions for the fungus through single factor experiment. Finally, we adopted L9(34) orthogonal test to optimize the components of the culture medium. The result showed that solubility starch was the optimal carbon source and soybean cake was the optimal nitrogen source .The optimal culture medium was solubility starch 5%, soybean cake 2%, KH2PO4 0.1%, MgSO4 0.05%, CaCI2 0.05%, GMP 0.03%, plant oil 1%. The optimal fermented condition: culture temperature 28℃, rotation speed 160r/min, pH6.5, the amount of liquid 70mL(250mL canonical flask), and fermentation time 7 days. 1.00g/100mL the initial mycelia production was increased to 1.76g/100mL. and the intracellular polysaccharide was increased by 53.68%.
