茶多酚对人羊膜上皮细胞FL系EROD活性的影响 被引量:4

摘要 本实验测定人羊膜上皮细胞FL系乙氧基异吩恶唑O-去乙基酶(EROD)的活性,观察茶多酚对FL细胞细胞色素P4501A1同功酶活性的影响。在茶多酚(GTPP)对猓被鼓芤种柒-NF对EROD的诱导作用。提示,茶多酚可能通过抑制细胞色素同功酶的活性,降低致癌物在体内代谢活化而发挥其化学预防肿瘤的作用。 The activities of ethoxyresorufin O-deethylase (EROD) was detected with Asokan′s method in cultured human amnion FL cells (FL cells) treated by different combination of green tea ployphenols (GTPP, 25,50 and 100 μ g·ml -1 ) and β -naphthoflavone ( β -NF,2×10 -5 mol·L -1 ). The resuls showed that the EROD activities were 9.50±1.55 trol),7.20±0.73,1.70±0.56 and 2.10±0.66(GTPP rteatment)(pmol resorufin·min -1 ·mg -1 protein)in experiment of inhibitoryeffect of GTPP on EROD inducing activity of control), 7.30±0.88,5.60±0.36,6.60±0.69 (GTPP control), 7.00±2.40,4.200.31 and 5.50±018(GTPP treatment)(pmol resorufin·min -1 ·min -1 protein) in experiment of inhibitory effect of GTPP on EROD activitis induced by β -NF. The results suggested that GTPP could inhibit either β NF induced activities of EROD and also interfere in the inducing effect of β -NF on EROD. GTPP (50) and 100 μ g·mol -1 ) could inhibit the constitutive activities of EROD in FL eclls. Thus the cancer preventive effect of GTPP might be due to its inhibitory effect on the metabolic activation of carcinogen by interfering in the expression and/of the enzymatic activities of cytochrome isozymes.
