摘要
应用32P后标记法检测人羊膜上皮细胞(FL)经不同浓度茶多酚(25μg/ml,50μg/ml和100μg/ml,终浓度,下同)和β-萘黄酮(2×l0-5mol/L)所形成的B(a)P-DNA加成物,观察茶多酚化学预防作用。结果表明,茶多酚不仅可以抑制β-萘黄酮对FL细胞细胞色素P450同功酶的活性的诱导,减少B(a)P-DNA加成物的形成;也可抑制经β-萘黄酮诱导后的FL细胞形成B(a)P-DNA加成物,而且该抑制效应与茶多酚剂量一致。茶多酚的化学预防作用可能与其影响细胞色素P450同功酶的表达调节以及抑制其活性有关。
With32Ppostlabeling method ,a study was performed to detect the formation of B(a)P-DNA adducts in cultured human amnion FL cells treated by different combinations ofgreen tea polyphenols(GTPP, 25μg/ml,50μg/ml and l00μg/ml )andβ-naphthoflaone(β-NF,Z×10-8mol/L ).The results showed that GTPP could inhibit either theβ-naph- thoflavone inducing expression of cytochrome P450 in FL cells,or the activity of the cy-tochrome P4s0 isozyme inβ-NF induced cells,thus reduce the formation of B(a)P-DNAadducts in these cells.
