摘要
研究了葡萄糖酶促磷酸化反应液中的二磷酸果糖经钙盐沉淀、草酸脱钙、树脂纯化和酒精-媒晶剂结晶法的工艺路线,通过单因素实验结合正交设计试验,优化工艺参数。试验研究表明,FDP钙盐的分离工艺参数为:pH=5.0,T=40℃,V(酒精)/V(水)=0.6,弱碱性阴离子RⅢ树脂在pH2.0吸附FDP,用0.05mol/LNaCl-0.01mol/LHCl洗脱无机磷等杂质,用0.5mol/LNaCl-0.1mol/LHCl洗脱FDP,FDP钠盐结晶工艺条件为pH4.5、温度25℃、媒晶剂10%、酒精加量1.6倍、结晶时间6.5h。FDP总收率达75%,重结晶体纯度达99.5%。
The technics route of glucose enzymatic phosphorylation to the fructose diphosphate in the fluid was studied in this article, through precipitation, the oxalic acid decalcify after the calcium salt, purification by the resin and the ethanol - intermediary crystalloid.Through the single factor experiment and the orthogonal design experiment, the optimized parameter of separating the FDP calcium salt is: PH 5.0, the temperature is 40 ℃, V (ethyl alcohol)/V (water) =0.6, the weak basicity anion R III resins adsorb FDP in pH 2.0, eluting impurity and inorganic phosphorus with 0.05 mol/L NaCl - 0.01 mol/L HCl, eluting FDP with 0.5 mol/L NaCl - 0.1 mol/L HCl, the crystallization condition of FDP sodium salt is: pH 4.5, the temperature is 25 ℃, crystalloid is 10 %, the ethanol is 1.6 times, the crystallization time is 6.5 hours. The receiving rate of FDP reaches 75 %, the purity of recrystallization reaches 99.5 %.
