骨髓间质干细胞诱导为肌样细胞分化相关基因的表达 被引量:8

摘要 目的:研究骨髓间质干细胞向肌样细胞分化前后骨骼肌特异性基因的表达。方法:体外分离成年SD大鼠骨髓干细胞,培养增殖,用5-氮杂胞苷诱导分化,RT-PCR检测分化前和分化后1d、2d、5d和7d骨骼肌特异性转录因子生肌决定因子MyoD、肌细胞生成素myogenin、MRF4以及肌肉特异性肌酸磷酸激酶MCK的表达。结果:MyoD在诱导前后均有表达,在诱导后1d表达显著上调(P<0·05);myogenin、MRF4和MCK在诱导前无表达,诱导后2d开始出现myogenin和MRF4的表达,诱导后7d开始出现MCK的表达。结论:骨髓间质干细胞能表达一定水平的骨骼肌细胞的分化调控基因MyoD,它向肌样细胞的分化可能与MyoD、myogenin和MRF4有关。 AIM: To analyze skeletal muscle specific gene expression in rat bone marrow mesenchymal stem cells (MSCs) differentiating into muscle- like cells. METHODS: The MSCs from adult SD rat were isolated, cultured and passaged in vitro by routine culture technique. Then, they were induced and differentiated with 5 - azacytidine. The expression of skeletal muscle specific transcription factors, MyoD, myogenin, MRF4 and muscle specific creatine kinase (MCK) were detected before and 1, 2, 5, 7 d after induction by reverse transcription polymerase chain reaction ( RT - PCR ). RESULTS : The expression of MyoD was detected before and after induction, its expression was markedly up - regulated 1 d after induction ( P 〈 0. 05 ). The expression of myogenin, MRF4 and MCK was not detected before induction. The expression of myogenin and MRF4 was detected 2 d after induction initially. The expression of MCK was detected 7 d after induction initially. CONCLUSION: MSCs can express skeletal muscle specific differentiating control gene MyoD to some extent. The differentiation of MSCs into muscle - like ceils may be related to MyoD, myogenin and MRF4.
