人类免疫缺陷病毒Ⅰ型慢病毒载体在脑梗死基因治疗中的应用 被引量:1

摘要 脑梗死的主要病理生理改变是能量耗竭、酸中毒、细胞内钙离子超载、氧自由基损伤、兴奋性氨基酸毒性、炎症细胞因子损害等。基因治疗则通过理想载体将目的基因导入脑组织内,通过目的基因的表达阻断上述病理生理过程,达到减轻缺血后脑损伤、促进神经功能恢复的目的。神经元是终末分化细胞,具有不可再生的特性。以神经元为靶细胞的基因治疗,对基因转移载体的要求更为严格。以人类免疫缺陷病毒Ⅰ型(HIV-Ⅰ)基因组为基础构建的HIV—Ⅰ慢病毒载体具有感染分裂细胞及非分裂细胞(包括神经元)的能力,
