胎盘绒毛血管生成状况及VEGF、MMP-2和MMP-9表达与早期自然流产的关系 被引量:27

Relationship between Angiogenesis,Expression of VEGF,MMP-2,-9 in Placental Villi and Spontaneous Abortion during Early Pregnancy

摘要 目的:探讨VEGF、MMP2和MMP9表达变化对胎盘绒毛血管生成的影响及与早期自然流产的关系。方法:选用CD34作为微血管标记物,应用免疫组化检测30例早期自然流产病例(自然流产组)和20例早期人工流产病例(人工流产组)胎盘绒毛微血管密度(MVD);RT-PCR检测胎盘绒毛VEGFmRNA的表达;酶谱分析法测定MMP-2和MMP-9的表达。结果:自然流产组VEGF121mRNA表达及MVD均明显低于人工流产组(P<0.01)。相关分析显示,两组中VEGF121mRNA相对表达量与MVD值均呈显著正相关(rθ=0.9794,rλ=0.9183)。自然流产组酶原型MMP-9的表达量显著低于人工流产组(P<0.01),激活型MMP-2的表达量显著高于人工流产组(P<0.01)。结论:孕期胎盘绒毛内VEGF121mRNA低表达所致的血管生成障碍以及MMP-2和MMP-9表达失调可能共同参与了早期自然流产的发生发展过程。 Objective: To explore relationship between angiogenesis, expression ofVEGF, MMP-2, -9 in human placental villi and early spontaneous abortion. Methods: Thirty women of spontaneous abortion during early pregnancy were included in study group, 20 women of induced abortion during early pregnancy were selected as control group. Microvessel density (MVD) in placental villi was measured by immunohistochemistry method by selecting CD34 as a marker ofmicrovessel. Expression level ofVEGF nRNA in placental villi was detected by RTPCR. Expression level of MMP-2, -9 protein in placental villi was measured by gelatin zymography technique. Results: Both expression of VEGF121 mRNA and MVD in spontaneous abortion group were lower than that in induced abortion group (P〈0.01), a positive correlation between expression of VEGF121 mRNA and the MVD can be showed in either spontaneous abortion group or control group (rθ=0.979, P〈0.001; rλ=0.918, P〈0.001). In spontaneous abortion group the expression of pro-MMP-9 decreased obviously (P〈0.01), and the expression of active MMP-2 increased significantly (P〈0.01), when compared with control group. Conclusion: The deficient angiogenesis caused by the lower expression of VEGF121 mRNA and the expressive maladjustment of MMP-2 and MMP-9 in placental villi may be concerned with spontaneous abortion during early pregnancy.
