摘要
用高压液相色谱法测定安神二号蜜丸制剂和赤芍中芍药甙的含量,以寻找控制制剂和药材质量的方法。安神二号蜜丸和赤芍饮片经冷浸、温浸、回流、超声处理,芍药甙提取完全。采用YWGC18不锈钢柱(150/4.6/10),以甲醇/0.05mol(pH3.6)磷酸二氢钾(35:65)为流动相,在流速0.8ml/min、工作压力22600kPa、检测波长240nm的条件下,芍药甙tR为6.10min,单一样本测定时间为12min。芍药甙在2~40μg/ml范围内线性关系良好,r=0.9998(n=5),回收率为98.03%,CV=0.44%(n=3)。日内误差为2.25%(n=6),日间误差为2.24%(n=6)。安神二号蜜丸和赤芍中芍药甙的含量均值分别为0.318%(n=3)和4.52%(n=3),CV分别为2.63%和2.31%。
Calcium dobesilate was synthesized by K2Cr2O7 oxidizition, K2S2O5 addition and Ca(ClO4)2 exchange reaction using p-Hydroquinone as begining material. The pure product was obtained through washing by Ph-Me-Ethol and recrystallization by hot water. No impurity was observed by chromatographic analysis. The purity is above 99%by HLCT determination. The chemical structure was confirmed by IR and element analysis.
