摘要
目的观察健康人外周血单个核细胞(PBMC)不同细胞亚群和活化的CD4+、CD8+T细胞上TIM-3mRNA的表达,探讨TIM-3mRNA的表达与T细胞功能的关系。方法应用实时定量RT-PCR(realtimeRT-PCR)技术,检测健康人PBMC不同亚群和CD4+、CD8+T细胞受刺激后TIM-3mRNA的表达。结果健康人PBMC中CD14+单核细胞、CD4+T细胞TIM-3mRNA的表达量较高(分别为0.84±0.088、0.64±0.11),CD4+T细胞受非特异性刺激后TIM-3mRNA表达明显升高(0.57±0.065vs0.85±0.080,t=18.07,P<0.0001)。结论TIM-3mRNA可能优先表达在健康人PBMCCD14+单核细胞、CD4+T细胞上,活化后的CD4+T细胞TIM-3mRNA的表达增多,提示TIM-3的表达与TH细胞的功能有关,也可能与单核细胞中某些产生TH1型细胞因子的亚群有关。
Objective To investigate the expression of T cell immunoglobulin mucin 3(TIM-3) genein different cell types isolated from peripheral blood mononuclear cell(PBMC) and in activated T cells in order to find the correlation of TIM-3 expression with function of T ceils. Methods Expressions of TIM-3 mRNA in different cell types and activated CD4^+ , CD8^+ T cells from a healthy subject were detected by real time RT-PCR. Results The major sources of TIM-3 mRNA in the PBMC were CD14^+ monocytes and CD4^+ T cells. TIM-3 mRNA was significantly up-regulated in CD4^+ T cells after non-specific stimulation with PMA/ionomycin. Conclusion TIM- 3 mRNA may be preferentially expressed in CD14^+ monocytes and CD4^+ T cells and up-regulated in activated CD4 ^+ T cells. These results suggest that the expression of TIM-3 mRNA is functionally associated with TH cells and may be correlated with a small subset of CD14^+ monocytes producing TH 1 cytokines.
