摘要
以初代培养获得的彩色马蹄莲(Zantedeschia hybrida)品种‘Parfait’无菌块茎芽为外植体,研究不定芽诱导增殖的培养条件。4因素3水平的正交实验结果表明,以无菌块茎芽在含20 g.L-1蔗糖、pH 5.8的B5培养基中以每天12 h光照条件进行培养,对不定芽的诱导效果最好。培养基中添加不同种类和浓度的细胞分裂素,诱导率均可达100%,其中以添加3 mg.L-16-BA对不定芽的增殖效果最好。
Zantedeschia hybrida ‘Parfait' tuber buds as explant were sterilized and gowns to research induction and proliferation of adventitious buds. The culture conditions (light period, sucrose concentration, culture medium type and pH) were selected by orthogonal experiment. The result showed that the optimal culture condition for inducing adventitious bud was B5 medium containing 20 g · L^-1 sucrose, pH 5.8 under 12 h · d^-1 light. To stimulate proliferation of adventitious buds, different types and concentrations of cytokinin were supplemented to media, it was indicated that the induction rate was increased to 100% after adding cytokinin to media, and with a best proliferation rate when adding 3.0 mg·L^-1 6-BA.
