摘要
以美洲大蠊(PeriplanetaamericanaL)中枢神经系统的乙酰胆碱酯酶(AChE)作为筛选模型,采用Gorun等改进的Ellman比色法,建立测定AChE比活力的条件并改进,使方法规范化,在国内外尚无报导.并且初步测定20个N-甲基取代苯基氨基甲酸脂类化合物对AChE抑制活力,其差异显著,可用于追踪、检测和评估生物活性.此法的优点是灵敏度高、重复性强、省药省时,可为定向合成靶标的抑制性物质和构效关系的研究提供信息和科学依据.
The present paper reports that, based on a comparison among different preparxations and an investigation of adequate reaction factors, a screening model with uniformed assaying method is set up with acetylcholinesterase(AChE) from the central nervous system of adult Perplaneta americana L. As an application of this model. twenty N-methyl substituted benzene carbamates are tested for their inhibitory activities on AChE, and their strctureactivity relation is analyzed. The reslts show that the screening model with AChE is sensitive, repetitive, labor-saving and sample-saving.
