摘要 目的探讨发育期慢性铅暴露对仔鼠学习记忆及海马细胞N-甲基D-天冬氨酸受体(NMDAR)亚单位ε1、ε2mRNA表达的影响.方法采用跳台法检测小鼠被动回避反应的学习能力;采用逆转录-聚合酶链法检测慢性铅暴露时仔鼠海马细胞的NMDAR亚单位ε1、ε2mRNA表达水平.结果慢性铅暴露可致小鼠被动回避反应学习记忆能力下降,与对照组比差异有统计学意义(P<0.05);海马细胞的NMDAR亚单位ε1mRNA表达下降(2~4周),ε2mRNA表达水平基本不变.结论铅致仔鼠被动回避反应的学习能力下降,与其致海马细胞的NMDAR亚单位ε1mRNA表达水平下降有关. Objective To explore the effect of chronic lead at developmental stages on passive - aviodance learning and memory in hippocampus of mice and its changes of mRNA expression of ε1 (NR2A) and ε2(NR2B) subunits of the N - methyl - D - aspartate receptor(NMDAR). Methods The ability of acquiring and maintaining passive - avoidance reaction were observed by jumping stand test in mice after chronic lead poison. RT - PCR techniques were applied to recognize either the mRNA expression of ε1 and ε2 subunits of the NMDAR in hippocampus exposure to low level of lead. Results Lead exposure significantly reduced with control group( P 〈 0.05), in the hippocampus, from 2 to 4 weeks, the ε1 subunit mRNA expression in Pb^2+ - exposed group exhibited a significantly decline compared with control group, but there were no corresponding dchanges in the ε2 subunit tuRN expression between Pb^2+ - exposed group and cntrol group. Conclusion Chronic lead exposure results in decrease of passiveavoidance reaction learning and memory and its effect may related to significant decline ε1 subunit mRNA expression hippocampus.
