摘要 目的探讨细粒棘球绦虫Eg95抗原在BCG中的表达,为囊型包虫病疫苗的开发利用提供依据。方法用热诱导方法诱导rBCG-Eg95疫苗,用SDS-PAGE和Westernblot鉴定所表达的蛋白。结果表达重组蛋白为可溶性蛋白,分子质量单位为16.5ku。Westernblot示重组蛋白具有抗原性,用薄层扫描Bio-RadQuantityone分析系统分析,表达效率为占BCG菌体总蛋白的12%。结论细粒棘球绦虫Eg95抗原在BCG中成功表达,为BCG发展为多价疫苗载体奠定了基础。 Objective To study the expression of the Echinococcus granulosus 95 (Eg95) antigen encoding gene in BCG. Methods Recombinant BCG-Eg95 vaccine was induced daily at 45℃ for 45 rain at the last there days, The product from BCG was analyzed by SDS-PAGE and Western blot, Results The expressed recombinant Eg95 was soluble and could be detected by SDS-PAGE about the molecular weight of 16.5 ku, and possessed antigenicity in Western blot . The expression efficiency of Eg95 was about 8 of the total bacterial protein in BCG, Conclusion The E. granulosus 95 antigen gene in BCG is successfully expressed. The study could provide basis to develop the BCG multivalent vaccine vectors.
