人源性表皮高色素纯黑素细胞的培养及其生物学特性 被引量:3

摘要 目的:探讨如何获得人源性表皮高色素性纯黑素细胞,并鉴定其生物学特性。方法:以包皮组织作为细胞来源,消化法获得表皮细胞悬液,用两种不同的培养基培养细胞,参照文献并结合本实验室设计的方法对细胞进行纯化,Dopa染色和S-100染色鉴定细胞来源,酪氨酸酶活性、生长曲线及MTT法观察细胞生长状态。结果:获得的黑素细胞色素颗粒较多,未见成纤维细胞和角质形成细胞污染,生物学特性观察显示:高色素性黑素细胞具有贴壁时间长、高酪氨酸酶活性和生长增殖慢的特点。结论:获得了大量人源性表皮高色素性纯黑素细胞。 Objective In order to obstain the pure hunman epidermal melanocytes isolated from darkly-pigmented neonatal foreskin (HEMn-DP) and identify the biology characters for the mechanism and treatment of vitiligo. Methods The melanocytes were isolated and purified by referring to the formerly records and binding our ideas. The two different media were used to culture the cells.The cultured cells were stained by S-100 and Dopa.The biology conditions of the cells were examined by tyrosinase activity,growth curve and MTT detection. Results The melanocytes obtained by this method contained many melanosomes and were free of fibroblasts and keratinocytes contamination, To comparing the common melanocytes,the biological characters of HEMn-DP are longer adherence,higher tyrosinase activity and lower proliferation ability. Conclusion The pure HEMn-DP can be attained by our method.
