摘要
目的 建立一种无血清传代培养黑素细胞的方法 ,并观察抗角蛋白自身抗体 (AK auto Ab)对无血清培养黑素细胞及角质形成细胞增殖的影响 .方法 应用角质形成细胞无血清培养基 (KC- SFM)添加牛垂体浸出液 (BPE)和 TPA传代培养黑素细胞 ,应用 KC- SFM添加 EGF和 BPE传代培养角质形成细胞 ,培养均至第 3代时于培养基中加入不同浓度的 AK auto Ab,用 MTT法观察 AK auto Ab对两种细胞增殖的影响 .结果 应用无血清培养基成功地传代培养了黑素细胞 ,MTT法结果显示 AK auto Ab各浓度组对培养黑素细胞的增殖无明显影响 ,但对角质形成细胞的增殖有显著抑制作用并有浓度依赖性 .结论 建立了一种无血清培养黑素细胞的方法 ,AK auto Ab对黑素细胞的增殖无抑制作用而对角质形成细胞有特异性抑制作用 .
AIM To observe the effect of anti keratin autoantibody (AK auto Ab) on proliferation of melanocytes and keratinocytes cultured with serum free medium. METHODS Human melanocytes were cultured with KC SFM supplemented with BPE and TPA while human keratinocytes were cultured with KC SFM in the presence of EGF and BPE. Effects of different titers of AK auto Ab in medium on proliferation of melanocytes and keratinocytes were observed by means of MTT. RESULTS AK auto Ab had no effects on melanocytes but markedly inhibited the growth of target cells, keratinocytes, in a dose dependent manner. CONCLUSION We develope a serum free approach to the cultivation of melanocytes. AK auto Ab can selectively inhibite proliferation of keratinocytes.
