小鼠胚胎与子宫单层上皮细胞共培养的研究 被引量:23

摘要 本文报道建立了小鼠胚胎与小鼠子宫单层上皮细胞体外共培养系统。结果揭示;小鼠胚胎与 子宫单层上皮细胞共培养可以促进胚胎的发育、粘附和扩展;如果培养液中加入 3、67 × 10-6mol/L 17β-雌二醇,可以显著提高胚胎在共培养系统中的发育率、粘附率和扩展率。以上结果表明:小鼠 胚胎与小鼠子宫单层上皮细胞共培养系统是研究胚泡着床机理较理想的研究手段。 Project supported by a grant-in-aid from the Rockefeller Foundation, New York. The objective of this study was to develop a system in which mouse embryos were cocultured with uterine epithelial cell monolayer. Blastocysts were developed from eight-cell embryos in vitro. Monolayer of uterine cells were obtained from the uterine of day-3 pregnant mouse. Experiment A: Blastocysts were placed randomly into one of the four treatments: Ham's F-10 culture medium supplemented with 0. 5 % bovine serum albumin(BSA) with or without steroid hormones [ progesterone(P4 ) or estradiol-17β(E2) or P4 + E2 ]. Experiment B: Blastocysts were assigned to the 4 treatment groups consisting of Ham' s F-10 + 0. 5 % BSA on a uterine epithelial cell monolayer with or without sterol d hormones. All cultures were grown in 24-well culture plates, at 37t in a humidified atmosphere of 5 % CO2 in air. The results indicated that when uterine epithelial cell monolayers were used in co-culture and supplemented with 3. 67 × 10 -6mol/L estradiol-17β, the uterine monolayer grew to con fluence and mouse blastocysts hatched from the zona pellucida. Attachment and outgrowth were observed, and more embryos tended to hatch in uterine epithelial cell monolayer with steroid hormones than in uterine epithelial cell monolayer only (P < 0. 01 ). This may be an optimal system for studying in vitro implantation of mouse embryos.
