摘要
本实验着重研究了着床期家兔子宫内膜刀豆球蛋白 A(ConA)结合糖蛋白的变化。以非离子去垢剂 Triton X-100提取子宫内膜蛋白,经 ConA 亲和电泳发现,子宫内膜 ConA 结合蛋白总量在着床期略高于未孕期。采用 ConA-Sepharose 4B 柱亲和层析分离 ConA 结合蛋白,经 SDS-PAGE 分析发现,在着床期家兔子宫内膜中有5种 ConA 结合蛋白存在,其表观分子量分别为162、67.5、62.5、89及35kD,其中前三者含量在着床期不同阶段变化不大,89及35kD 蛋白以妊娠第六天含量最多。结果显示子宫内膜 ConA 结合蛋白在着床期存在阶段特异性变化。
The study was focused on the changes of the ConA binding protien in rabbit endometri- um during peri-implantation stage.It was found that the total amount of ConA binding protien in peri- implantation stage endometrium was higher than that during nonpregnant period with ConA affinity electrophoresis.By further isolation with ConA-Sepharose 4B column affinity chromatography and anal- ysis by SDS-PAGE,5 ConA binding protiens were identified and their apparent molecular weights were 162、67.5、62.5、89and 35kD,among which the amount of former three nearly had no changes in different periods of peri-implantation stage,whereas 89and 35kD protein were maximium on pregnant day 6.The results showed that the ConA binding protein of rabbit endometrium had a stage specific changes during peri-implantation stage.
