摘要
研究了单克隆免疫亲和柱-高效液相色谱法测定酒中赭曲霉毒素A的方法。脱气后的酒类样品用1%PEG和5%NaHCO3的水溶液稀释,然后通过Ochra Test免疫亲和柱净化,净化液经Hypersil BDS C18柱反相色谱柱分离、荧光检测器检测、外标法定量。对添加不同含量的赭曲霉毒素A进行6次反复实验,得其平均回收率为92.8%-99.5%,RSD%〈10,最低检测限为0.04ng/mL,线性范围为0.1-10ng/mL。
A method was developed for the determination of ochratoxin, （OA） in wine by immunoaffinity column clean - up and performance liquid chromatography. Degssing was performed to preventfast foam, 10ml of degassed wine wete diluted with 10ml of a water solution containing 1% PEG and 5% NaHCO3, then were cleaned up through OchraTest column, The separation was performed on a Hypersil BDS C18 column and acetonitrile - water（50：50） - 1% acetic acid was mobilephase . The linear range of the method was from0.1 - 10ng/mL and correlation coefficient was 0.9987.The quantitation limit was 0.04ng/mL. The recoveries from wine spiked with OA at levels ranging from 0.1 - 10 ng/mL were 92.8 - 99.5 %. and the linit of detection of the method was 0.04ng/mL.
