摘要
研究了单克隆免疫亲和柱 -高效液相色谱法测定牛乳中黄曲霉毒素M1的方法。样品通过离心脱脂 ,脱脂乳用免疫亲和柱净化 ,SpherisorMC18色谱柱分离 ,荧光检测器检测 ,外标法定量。对添加不同含量水平的黄曲霉毒素M15次重复实验的平均回收率为 80 .4 %～ 88.7%。变异系数(CV)为 7.7%～ 8.9%。检测低限为 0 .0 5μg/kg。
A method for determination of Aflatoxins M 1 in milk by High Performance Liquid Chromatography is reported. The skim milk sample is purified by AflaTest affinity cartridges. The Aflatoxins M 1 is separated on SpherisorM C 18 column, and is detected using fluorescent detector and is quantified by external standard procedure. Samples are added with Aflatoxins M 1 at the two levels. Five replicate analysis are performed at each level. The recoveries of the Aflatoxins M 1 are 80.4%～88.7%,and the variation coefficients are 7.7%～8.9%,the detection limits are 0.05?μg/kg。
