IL-12、IL-18基因对HIV-1外膜蛋白基因疫苗诱导的免疫应答的影响 被引量:3

摘要 目的研究IL-12、IL-18基因对HIV-1外膜蛋白基因疫苗诱导免疫应答的影响,以探求HIV-1核酸疫苗的新策略。方法pVAX1GP120联合IL-12、IL-18基因或者pVAX1GP120单独免疫BALB/c小鼠,采用ELISA检测免疫小鼠的特异性抗体和IFN-γ水平,以MTT比色法检测免疫小鼠脾淋巴细胞的增殖,用乳酸脱氢酶(LDH)试验检测小鼠特异性细胞毒性T淋巴细胞(CTL)的应答。结果与pVAX1GP120免疫组比较,pVAX1GP120联合IL-12、IL-18基因免疫组小鼠血清的抗HIV-1gp120抗体滴度升高,IFN-γ升高,小鼠的脾淋巴细胞增殖实验刺激指数(SI)以及特异性CTL活性均高,差异均有统计学意义(P<0.01)。结论IL-12、IL-18基因联合HIV-1核酸疫苗免疫小鼠,有可能增强特异性TH1细胞和CTL反应,IL-12、IL-18基因对体液免疫可能有抑制作用。因此,IL-12、IL-18基因对于治疗性HIV-1核酸疫苗可能是具有较好应用前景的免疫佐剂。 Objective To investigate the effect of IL-12, IL-18 gene immunization on immune response induced by HIV-1 co-administered envelope nucleic acid vaccine(DNA vaccine) and to explore new strategies for HIV-1 envelope nucleic acid vaccine. Methods BALB/c mice were immunized with pVAX1GP120 alone or with the DNA encoding for IL-12, IL-18. Their sera were collected for analyz/ng anti-HIV antibody and IFN-γ by ELISA, and splenocytes were isolated for detecting antigen-specific lymphoproliferative responses and specific CTL response by MTT assay and LDH assay respectively, Results The anti-HIV antibody titers of mice co-immunized with pVAX1GP120 and the DNA encoding for IL-12, IL-18 were lower than that of mice immunized with pVAX1GP120 alone(P 〈 0.01). The IFN-γ level of mice co-immunized with pVAX1GP120 and the DNA encoding for IL-12, IL-18 was higher than that of mice immunized with pVAX1GP120 alone( P 〈 0.01 ). Furthermore, compared with mice injected with pVAX1GP120 alone, the specific CIL cytotoxity activity and antigen-spocific lymphoproliferative responses of mice immunized with pVAX1GP120 and the DNA encoding for IL-12, IL-18 were significantly enhanced respectively (P〈0.01). Conclusion The DNA encoding for IL-12, IL-18 together with HIV DNA vaccine may enhance specific TH1 responses and cellular immune responses elicited in mice. Moreover, the DNA encoding for IL-12, IL-18 may down-regulate the humoral responses. Hence the DNA encoding for IL- 12, IL-18 are potential immune adjuvants for therapeutic HIV DNA vaccine.
