Experimental study of the changes of gap junction in unstable bladder

摘要 目的研究不稳定膀胱逼尿肌细胞间间隙连接蛋白(Cx)的改变,探讨其与不稳定膀胱发病机理的相关性。方法建立Wistar大鼠膀胱出口梗阻模型,行充盈性膀胱测压确定不稳定膀胱(USB)组13只,另设对照组10只。均为健康Wistar雌性大鼠,体重200~220g。采用Westernblot技术、双重标记的荧光免疫组织化学及激光共聚焦显微镜技术定量分析逼尿肌Cx43含量改变。结果Cx43蛋白条带在USB组和对照组均有表达,相对分子质量为43000。USB组逼尿肌蛋白条带平均灰度值为31.066,对照组为11.701,2组差异有统计学意义(P<0.01)。激光共聚焦扫描观察Cx43表达主要出现在细胞与细胞连接处。USB组Cx43像素密度为29.5250±13.9176,荧光斑面积为(17.9290±8.8403)μm2,对照组Cx43像素密度为14.2140±2.1747,荧光斑面积为(5.7480±3.1075)μm2;两组比较差异均有统计学意义(P<0.05)。结论逼尿肌细胞间间隙连接蛋白增多引起兴奋传递的增强可能是不稳定膀胱发病的重要因素之一。 Objective To study the quantitative changes of gap junction (GJ) in the detrusors of unstable bladder(USB) in rats, and to deduce the functional changes of GJ, which mediates intercellular communication,so as to demonstrate one of the mechanisms of USB. Methods Thirteen Wistar rats, which were identified to have USB by manometry of filling bladder after establishment of bladder outlet obstruction (BOO) model, served as study,ie, USB group. Another 10 healthy female Wistar rats served as control group. The content and distribution of connexin (Cx) 43 of the detrusors, which were taken from these rats ,were quantitatively analyzed by Western blot and laser confocal microscopy with a double-label immunohistochemical technique. Results Cx43 protein was expressed in both control and USB group, the relative molecular weight was 43000. The mean gray levels of the detrusor protein bands in USB group and control group were 31. 066 and 11. 701, respectively; the difference of the values between the 2 groups was significant (P 〈 0.01 ). The expression of Cx43 was found mainly in the intercellular junction of the detrusor with laser confocal microscopy;and the Cx43 increased obviously in USB group [ pixel density,29. 5250±13.9176;staining size ( 17. 9290 ±8. 8403 )μm^2] compared with those in control group [ pixel density, 14.2140±2. 1747; staining size, (5.7480 ±3. 1075) μm^2 ;P 〈0.05].Conclusions The increase of Cx in GJ of the detrusor,which leads to an enhanced intercellular excitatory communication, may be one of the important mechanisms of unstable bladder.
