摘要
以40多种不同pH值的甲醇-水体系作为流动相,在10多种ODS柱上对212种药物进行了高效液相色谱法(HPLC)的色谱行为测试,以保留值为流动相pH值的函数S(pn)特征作指标,将所测试的药物分成碱性、酸性和中性三类。分类有利于从宏观上把握药物的色谱行为规律,减少不必要的实验条件和实验次数,对内标的选择和分离度的改善均有指导意义。
In this paper the retention behaviours of about 200 drugs were investigated under reversed-phase conditions. Experiments were performed with more than 40 mobile phases contoining 0.1% triethylamine and 10. 0% -80. 0% methanol with PH values ranging from 2. 0 to 6.0. The retention values of these drugs were measured on several columns including LiChrosorb RP-18, Shim-pack ODS, Nucleosil C_(18), YWG-C_(18), NovaPak, μ Bondapak and Zorbax ODS etc. The new parameters, S_p, S_(pn), indicating retention values as the function of mobile phase PH were presented. In terms of the characterization of S_(pn), the 212 drugs were divided into three classes: basic ( S_(pn)>0.1), acidic(S_(pn)< - 0.1), and neutral drugs (-0.1< S_(pn)<0.1). The basic drugs range as high as over 60% of all drugs. The PH- S. graph can distinctly illustrate the effective power and direction of action of the eluent PH value on different drugs.For most basic drugs, the variation of retention values are greater and more sensitive to the eluent pH value than for acidic and neutral drugs. Chromatographic optimization for basic drugs is more difficult than for others. The reversal of elution order is frequently encountered with basic drugs between PH 4 and 5. The phenomenon is defined as 'the elution order reversal region of PH 4-5 for basic drugs'. Experimental data showed that the retention behaviour of a drug was not absolutly in association with its pKa value.This classification might play important roles in the recognition of the trends of chromatographic behaviour of these drugs, the determination of initial chromatographic condition and the reduction of unneccessary optimum factors. It may be helpful for us to obtain the optimum separation results and to select the best internal standards from all these drugs in pharmaceutical analysis.
