摘要
采用反相高效液相色谱法,在色谱行为数据库技术的指导下,快速地建立了以甲醇- 磷酸缓冲盐(30∶70,pH2.5)为流动相,环丙沙星为内标,三氯乙酸为蛋白沉淀剂的样品预处理方法,对人血清中的氟罗沙星浓度进行了测定。
A method to determine the serum concentration of fleroxacin with high performance liquid chromatography was established within 3 to 4 days with the assistance of chromatographic behavior database. The chromatography includes a LC 4A liquid chromatograph and a spectrophotometric detector SPD 2AS (Shimadzu, Japan). A μ Bondapak C 18 , 10μm column was used as solid phase. Mobile phase was prepared by adding methanol to water (30∶70), pH2.5, containing 0.1% TEA. Ciprofloxacin (CPLX) was selected as inner standard. Serum samples were pretreated with 10% TCA to remove protein. To the result, the limit of detection was 0.1mg/L. The peak height ratio of FLRX to inner standard correlated well with the concentration of FLRX in the range of 0.125 to 4.0mg/L ( r =0.9999). The average inner and inter day coefficient variance were 4.1% and 5.1% respectively.
