摘要
目的:考察不同提取方法对胡芦巴碱提取率的影响,建立胡芦巴中胡芦巴碱含量测定的高效液相色谱方法.方法:分别以水、甲醇、无水乙醇及三种比例的甲醇-水为溶剂,用超声等方法考察提取率.HPLC测定采用Zorbax XDB-C18分析柱,流动相为0.37mmol·L-1磷酸溶液(pH=3.55),检测波长265nm.结果:以甲醇-水(1:1)超声提取40min胡芦巴碱的提取率较高;样品中胡芦巴碱进样量在0.40~3.96μg范围内线性关系良好,r=0.9999(n=5),加样回收率98.18%,RSD=1.54%(n=6);33份胡芦巴商品药材中胡芦巴碱的含量为0.243%~0.731%,超过文献报道的胡芦巴碱含量1倍以上.结论:提取及含测方法操作简便,结果可靠,可为胡芦巴质量控制提供科学依据.
Objective : To develop a method for the extraction and determination of trigonelline in Huluba (seeds of Trigonella foenum-graecum L. )and compare the effect of different extraction methods by determination of trigonelline. Metbod:Huluba sample was extracted by ultrasonic cleaner(500W)and other methods in methanol,absolute ethanol,water and different concentrations of methanol-water,respectively. The experimental conditions were as follows,the Angilent Zorbax Eclipse XDB- C18 column,mobile phase 0. 37 mmol · L^-1 phosphoric acid (pH= 3.55),flow rate 1 ml· min^-1 ,and detection wavelength UV 265nm. Results:In the extraction with ultrasonic cleaner (500W) in methanol-water(1 : 1),higher efficiency was showed. The standard curve was linear in the range of 0.40- 3.96μg ,r = 0. 9999 (n = 5). The recovery rate was 98.18 %, RSD: 1.54 % (n= 6 ). The result of determination of trigonelline in 33 comercial samples of Huluba was 0. 243 % -0.731% which was more than two times its value reported in literatures. Conclusion:A method for extraction and determination of trigonelline was established ,which was efficient ,simple and reliable ,and can be used for the quality control of Huluba(fenugreek).
