摘要
目的:评价大青叶不同化学部位抗单纯疱疹病毒Ⅰ型的活性.方法:利用Hep-2细胞体外培养系统,通过噻唑蓝(MTT)比色法和观察细胞病变效应(CPE),以治疗指数(TI)为评价指标,评价大青叶不同化学部位体外抑制单纯疱疹病毒Ⅰ型(HSV-Ⅰ)对Hep-2细胞的感染作用;并通过改变加药方式,初步探讨抗病毒机制.选择体外抗病毒活性较强的部位,观察其对HSV-Ⅰ脑炎小鼠的保护作用,以综合评价其抗病毒活性.结果:大青叶Ⅱ~Ⅴ部位对HSV-Ⅰ有直接灭活作用,各部位均不能阻止HSV-Ⅰ侵入细胞,除Ⅲ,Ⅴ部位外,其余部位均有抑制HSV-Ⅰ生物合成作用;Ⅳ部位能显著降低HSV-Ⅰ脑炎小鼠死亡率.结论:大青叶Ⅳ部位具有较强的体内外抗HSV-Ⅰ病毒活性.
Objective: To evaluate the antiviral effect of different chemical fractions isolated from Folium Isatidis on herpes simplex virus type Ⅰ (HSV-Ⅰ). Method: Anti-HSV-Ⅰ effects of Folium Isatidis were investigated in Hep-2 cell by adopting MTr colorimetric assay and observing cytopathic effect (CPE). Treatment index (TI) was used to evaluate the antiviral activity of different chemical fractions from Folium Isatidis. The antiviral mechanism of Folium Isatidis was investigated by changing the different ways of drug administration. Result: The fractions Ⅱ-Ⅴ from Folium Isatidis inactivate HSV-Ⅰ directly. None of the chemical fractions had antiviral effects of adsorption-blocking. The chemical fractions except fraction Ⅲ and fraction Ⅴ inhibited the biological synthesis of HSV-Ⅰ. The fraction IV significantly reduce death rate of mice infected with HSV-Ⅰ .Conclusion: The fraction Ⅳ from Folium Isatidis has powerful anti-HSV-Ⅰ effect in vitro and in vivo
