摘要
目的观察全反式维甲酸(ATRA)和顺铂(CDDP)在体外对人卵巢癌细胞株3AO的增殖、分化和凋亡的影响,探讨全反式维甲酸提高3AO细胞株对顺铂化疗敏感性的机制。方法ATRA,CDDP以及两药联合分别处理卵巢癌3AO细胞株,MTT法检测细胞存活率,瑞氏染色后高倍镜下计数凋亡细胞数,细胞免疫化学测定bcl-2,p53的表达。显微镜下观察细胞形态变化。结果细胞存活率在联用组中随ATRA剂量的增加而下降,且低于同剂量单用组(P<0·01);细胞凋亡率在联用组中随ATRA剂量的增加而增加,且高于同剂量单用组(P<0·01);bcl-2在联用组中随ATRA剂量的增加而下降,p53在联用组中随ATRA剂量的增加而增加。细胞形态随ATRA剂量的增加而趋向于成熟细胞。结论全反式维甲酸能抑制卵巢癌细胞株3AO的增殖,诱导其分化,提高其对顺铂促凋亡效应的敏感性。其作用机制可能与bcl-2的表达下降有关,p53作用尚不清楚。
Objective To explore the effects of all- trans retinoie acid (ATRA) and eisplatin (DDP) on the proliferation, differentiation and apoptosis of ovarian carcinoma cell line 3AO in vitro and the potential mechanism of increased response rate of CDDP induced by ATRA. Methods Human ovarian carcinoma cell line, 3AO, was respectively treated by ATRA, DDP and ATRA combined with DDP. Growth inhibition was measured by MTT assay, and the index of apoptosis was evaluated by Wright' s staining. The expressions of bcl - 2 and p53 were analyzed immtmohistochemieally. The changes of cell morphology were observed under microscope. Results In ATRA + DDP group, the survival rate of 3AO cells decreased more than ATRA group ( P 〈 0.01 ) or DDP group ( P 〈 0.01 ). The effects of growth inhibition are independent on the concentration of ATRA. In ATRA + DDP group, the index of apoptosis increased more than ATRA group ( P 〈 0.01 ) or DDP group ( P 〈 0.01 ). With the increase of ATRA concentration, the index of apoptosis increased in ATRA + DDP group, but not in ATRA group. In ATRA + DDP group, the expression of bcl - 2 decreased anti p53 increased with the increases of ATRA concentration. The cell differentiation was observed with the ATRA increasing. Conclusions ATRA can suppress the cell proliferation, induce cell apoptosis, and improve the sensitivity of 3AO cells to DDP. The possible mechanism may involve the change of bcl - 2, yet the role of p53 is unclear.
