摘要
目的:研究在血小板源性生长因子(PDGF)和白介素-1β(IL-1β)作用下苏拉明(suramin)对培养的人视网膜色素上皮(retinapligmentepithel-um,RPE)细胞增殖的影响。方法:将不同浓度的(15,150,250mg/L)苏拉明分别加入用PDGF10μg/L或IL-110μg/L培养的RPE细胞培养液中,继续培养3d后采用四甲基偶氮唑盐(tetrazoliu,mMTT)比色法,细胞分裂指数计数和核仁组成区嗜银染色(AgNORs)检测在细胞因子作用下不同浓度苏拉明对RPE增殖活力的影响。结果:含有PDGF10μg/L或IL-1β10μg/L的培养液显著刺激RPE的增殖,苏拉明明显抑制了这两种细胞因子条件下RPE的增殖,在150mg/L时的抑制率分别为26%和18%,对细胞的形态无明显影响。结论:苏拉明对PDGF或IL-1β刺激的RPE的增殖有显著抑制作用,该作用为非毒性作用。进一步证明苏拉明对增生性玻璃体视网膜病变(prolifera-tivevitroeretinopath,PyVR)中相关细胞因子的拮抗作用,为临床防治PVR提供了新的用药思路。
AIM: With the effect of PDGF and IL-1, investigate effects of suramin on proliferation of cultured human retinal pigment epithelial (RPE) cells in vitro. METHODS: The human RPE cells cultured with PDGF or IL-1 were added with suramin (15,150,250mg/L). Three days later, cells were measured in tetrazolium colormetric assay (MTT), cell division index and AgNORs staining. The effect of suramin in different concentration to proliferation of RPE under contribution of cytokine were examined. RESULTS: The PDGF 10μg/L or IL-I 10μg/L stimulated RPE proliferation significantly. The suramin could effectively inhibited the proliferation caused by these cytokine. At the concentrations of 150 mg/L, suramin's inhibition rate were 26% and 18% in PDGF and IL-1 group respectively. CONCLUSION: The PDGF or IL-1 induced proliferation of RPE can be effectively inhibited by suramin. It's not toxic effect. It proved that suramin can antagonistic the effect of cytokine in proliferative vitreoretinopathy (PVR). It's useful in treatment of PVR.
