摘要
检测地塞米松对体外培养的人视网膜色素上皮细胞生长的影响.对hRPE细胞进行常规培养,取3-4代生长良好的细胞用于实验,采用MTT法研究不同浓度、不同作用时间,地塞米松对hRPE细胞增殖的影响.用免疫细胞化学染色法检测添加不同浓度、不同作用时间地塞米松对hRPE细胞核内细胞增殖核抗原表达的影响.MTT法发现小剂量地塞米松促进hRPE细胞的增殖,50-200 mg/L浓度组随着浓度的增高,hRPE细胞的增殖更加明显.大剂量地塞米松抑制hRPE细胞的增殖,且随着浓度的增高,对hRPE细胞的增殖的抑制更加明显.免疫细胞化学染色法发现,在50-200 mg/L浓度组,随着浓度的增高,hRPE细胞核内PCNA表达增强,光密度值增大,在400-800 mg/L浓度组,随着浓度的增高,hRPE细胞核内PCNA表达减弱,光密度值减少.小剂量地塞米松促进体外培养的hRPE细胞的增殖及细胞核内PCNA的表达,大剂量地塞米松抑制体外培养的hRPE细胞的增殖及细胞核内PCNA的表达.
This paper investigates the effect of Dexamethasone (DEX) on the proliferation of human retinal pigment epithelium (hRPE). The hRPE cells were cultivated in conventional method, and 3-4 generations of were used for the tests. The proliferation of hRPE treated by DEX in different concentration and different action time was tested with MTF assay. Cytochemieal staining method was used to detect the expression of proliferating cell nuclear antigen in hRPE treated with DEX in different concentration and different action time. Results show that the primary human RPE cells were hexagonal, and that the cytoplasm was filled with melanin granules, subcultured cells, short spindle or a spindle-shaped particulate little or none. As MTT assay shows, DEX could increase cells proliferation in low concentration, and that 50-200 mg/L, the optical density and the number of hRPE cells increased with the addi- tion of the concentration of DEX in 50-200 mg/L, DEX could decrease cells proliferation in high concentration. DEX in 400-800 mg/L could decrease cells proliferation and the optical density and the number of hRPE cells decreased with the addition of the concentration of DEX. Cytochemical staining method shows that the PCNA expression of hRPE cells and optical density increased with the addition of the concentration of DEX in 50-200 mg/L, and the PCNA expression of hRPE cells and optical density decreased with the addition of the concentration of DEX in 400-800 mg/L.
