摘要
目的:观察染料木黄酮(genistein,Geni)对体外培养的兔结膜成纤维细胞(rabbitconjunctivalfibroblasts,RCF)增殖的影响。方法:应用MTT法测定不同生长时间及浓度的Geni,5-FU及二者联合药物对RCF生长的影响。结果:2.5 ̄40mg/LGeni对RCF生长抑制率分别为27.2%,34.0%,44.6%,55.1%,58.9%;不同浓度的Geni作用1,3,5,7d的生长抑制率,随时间、剂量增加而加大。Geni(5,10mg/L)与5-FU(1mg/L)联用的抑制率分别为45.6%和49.1%。结论:Geni对RCF的增殖有抑制作用,呈时间和剂量依赖性;与5-FU有协同效应。
AIM: To investigate the effects of genistein on the proliferation of rabbit conjunctival fibroblasts (RCF) in vitro.
METHODS: Cultured RCF were exposed to different concentration and time of genistein, 5-FU and genistein combined with 5-FU. The proliferation of fibmblasts were investigated by MTT assay. RESULTS: Genistein (2.5,5,10,20,40mg/L) suppressed the proliferation of RCF in vitro .The rate of inhibition were 27.2% ,34.0% ,44.6% ,55.1% ,58.9% respectively. The inhibition rate showed a significant difference at different time points (1,3,5,7d) of exposure to genistein. When genistein (5,10mg/L) combined with 5-FU (lmg/L), the rate of inhibition were 45.6% and 49.1% respectively.
CONCLUSION: Genistein has a significant inhibitory on effect on the proliferation of RCF. The effect is in a dose and time-dependent manner. 5-FU and genistein have synergistic effect on inhibiting proliferation.
