摘要
从一支保存多年的金黄色葡萄球菌RN 1304(rec^-)的冷冻干燥菌种管中分离得黄色(Y)菌株及其白色(W)变株,它们都具有Tc^r表型及一条质粒带。通过SDS处理。消除了它们的Tc^r表型及质粒带,获得质粒消除株RN1304(Y-1)(Tc^s)及RN1304(W-1)(Tc^s)将其作为受体菌,二株临床分离的金黄色葡萄球菌多剂耐药株(82022及85030)作为供体菌,通过葡萄球菌噬菌体80进行Tc^r质粒的转导,证明二株质粒消除菌能在葡萄球菌的质粒转移中作为有效的受体菌,它们的转导频率为10^(-7)_10^(-8)。通过转导,同时鉴定了金黄色葡萄球菌82022及85030都具有Tc^r质粒。质粒大小与原始RN1304株及已知金黄色葡萄球菌(22)的Tc^r质粒相近似。
Staphylococcus aureus RN1304(rec^-)(Y)and its albus variant(W) were isolated in an old freeze dry ing culture. Both strains showed Tc^r phenotype and a plasmid band in agarose get electrophoresis.Strains of Tc^r plasmid eliminated were obtained by treated Y and W strains described above with SDS. Two plasmid eliminated strains: RN1304(Y-1)(Tc^s) and RN1304(W-1)(Tc^s)were used as recipients, two multiple drug resistant strains of S. aureus.82022 and 85030 from clinical source as donors,and staphylococcal phage 80 as transducing phage, Tc^r plasmid transduction was carried out.By trans-duction it was demonstrated that strains of Y-1 and W-1 were capable of serving as recipients in genetic transfer of staphylococcal plasmid Their transduction frequency was similar, about 10~7—10~8.By transduction the Tc^r plasmids of S.aureus 82022 and 85030 were also identified
