生米卡链霉菌与北里链霉菌种间原生质体融合重组的研究 被引量:1

STUDY ON PROTOPLAST FUSION BETWEEN STREPTOMYCES MYCAROFACIENS AND STREPTOMYCES KITASATOENSIS

摘要 生米卡链霉菌和北里链霉菌用42%PEG4000做助融剂进行多配对原生质体种间融合,融合率为10^(-2)。同时,进行了UV灭活亲株的种间融合,融合率也为10^(-2)。采用液体培养基再生,表观融合率达10^(-1)。随机挑选形态各异的一定数量的融合子作产物分析,发现它们产生柱晶白霉素,总效价比直接亲本高5.9倍,比间接亲本高27%。高效液相色谱测定表明A_5组份从9.1%提高到43.4%。 Interspecies fusion of several mating pairs were carried out with 42% PEG4000 as fusion agents, the fusion rates were 10^(-2). Meantime, we undertook the fusion with the inactivated protoplasts of parents strains by UV, the fusion rates were also 10^(-2). By means of liquid medium regeneration, the 'apparent' fusion freq uencies were 10^(-1) Varied forms of recombinants with a definite quantity by random were selected and analysed. The results showed that it was Leucomycin and the total output was 5.9 times higher than that of direct parent and 27% than that of indirect parent The analysis on components by Craig countercurrent Apparatus indicated that component A5 has increased from 9.1% to 43.4%.
