摘要
新分离的一株液化沙雷氏菌(Serratialiquefaciens)能产生大量胞外碱性脂肪酶,本文在摇瓶发酵水平上考察了培养基组成、培养条件等因素对其生成脂肪酶的影响。在优选条件下,即培养基组成为(%)玉米油1.25,豆饼粉2.0,蛋白胨1.0,酵母膏0.2,K_2HPO_40.2,MgSO_4·7H_2O0.1,初始pH为7.25时,28℃,150r/min旋转摇床振荡培养40h,该菌株的发酵液酶活达到43u/ml,比优化前产酶量(17u/ml)提高近两倍。关于液化沙雷氏菌胞外脂肪酶的研究,国内外目前均未见报道。
Serratia liquefaciens strain S33DB-1 was capable of producing large amountof extracellular alkaline lipase under the inducement of oils.Factors affecting the lipaseproducti on were investigated in shaken-flask level.Obtained results showed that theoptimum medium composition and cultural conditions were(%):Bean cake meal 2.0,Cornoil 1.25, Peptone 1.0, Yeast extract 0.2,K_2HPO_4 0.2, MgSO_4·7H_2O 0.1 ,pH7.25,and 28℃,25ml medium in a 250ml flask,150r/min shaking for 40h.Under such conditions, thelipase activity in culture supernatent was 43u/ml,as 2 times high as the original one.
