摘要
从生霉棉布上分离出的康宁木霉CP88105经诱变处理,获得一株高产纤维素酶的突变株CP88329。在固体培养基上,28℃培养72小时,所产纤维素酶固体曲,以羧甲基纤维素钠为底物酶活力为4880u/g;以脱脂棉为底物酶活力为480u/g。产酶活力水平均为出发菌的3倍。酶在脱脂棉上作用最适条件为pH4.5-5.0,45-50℃;45℃保温4h,pH稳定范围为3.5-6.5;60℃保温1h,酶活力剩余20%。酶可用于苎麻布抛光处理和牛仔服“石磨”处理。
A mutant strain CP88329 with high potency for cellulase prodution was obtained fromTrichoderma ningii CP88105 which was isolated from contaminated cotton by treatment at 80℃for 10 min,and then with 0.04%of N-methyl-N-nitro-N-nitrosoguanidin at room temperature for30min。Cellulase kojiwas obtained by growing the mutant strain on semi-solid medium consisted of3g of rice straw powder,2g of wheat bran and 10 ml of 1%ammonium sulfate in 250 ml of flasksat 28 ℃ for 72 h at initial pH 6.0.Enzyme activity of the koji was 4880 u/g on carboxymethylcellulose-Na and 480 u/g on cotton。Cellulase activity of the koji from the mutant strain wasabout 3 times that of the parent strain。The optimal conditions for enzyme action on cotton were pH 4.5-5.0 and 45一50℃。Theenzyme was stable at pH 3.5-6.5 at 45℃ for 4h.After incubating the enzyme at 60℃ for 1 h,20%of the enzyme activity remained。The cellulase has been used for polishing treatment of rame cloth and“stone polish”treatment of cowboy clothes。
