摘要
通过研究神经节苷脂GM_3对国人单核样白血病细胞系J6-2细胞蛋白质磷酸化的影响,在[γ- ̄32P]ATP,GM_3,ATP,Mg ̄(2+)与J6-2细胞液及颗粒两部分共同反应,1Omin(30℃)体系中,观察到GM_3对两部分蛋白质磷酸化的调节作用。GM_3(100μmol/L)促进颗粒部分分子量为180000,87000,78000,67000,43000及31000的蛋白质磷酸化,促进胞液部分分子量为87000及56000的蛋白质磷酸化,而且能抑制70000及43000蛋白质磷酸化。由于GM_3已被前人证实能对J6-2细胞起分化作用,其作用时间长达4-6d,很可能GM_3对蛋白质磷酸化作用的调节是GM_3促分化作用的早期信号。
The effect of ganglioside GM_3 onthe endogenous protein phosphorylation of hu-man monocytoid leukemia cell line J6-2 cellswas studied.Addition of 100 μmol/L GM_3 tothe particulate fraction of J6-2 cells enhancedthe endogenous phosphorylation of severalproteins whose relative molecular mass were180,87,78,67,43 and 31kD while additionof 100 μmol/L GM_3 to the cytosol fraction of16-2 cells stimulated the phosphorylation ofseveral proteins whose relative molecular masswere 87 and 56kD,but could inhibit the phos-phorylation of 70 and 43kD proteins。BecauseGM_3 has been reported to be able to induce J6-2 cells to differentiate along the monocyte/macrophage route by long term incubationwith GM_3(up to six days),so the authorspostulated that protein phosphorylation in-duced by short term GM_3 treatment might bethe early signal of differentiation of J6-2 cells.
