摘要
用碱性裂解法从麦迪霉素产生菌(Str.mycarofaciens)10204中分离到质粒DNA,经琼脂糖凝胶电泳分析和电镜观察得到证实,分子量为11.5kb(7.6×10~6道尔顿),名为pSMVI。该质粒经酶切分析表明具有4个Bam HI位点和1个EcoRI位点,用质粒消除和转化实验,说明该质粒与麦迪霉素产生有一定的关系。
The isolation and some properties of plasmid DNA in StrePto-myces mycarofaciens var. Sichuanensis producing Midecamycin were studied.The plasmid DNA in Str. mycarofaciens 10204 was isolated by means of the alkaline lysis method. Plasmid DNA bands were observed with agarose gel electrophoresis; circular plasmid DNAs were also confirmed with electron microscope. The molecular weight of the plasmid,named pSMVI,was 11. 5kb(7.6×106 daltous). The DNA was digested into four fragments with BamHI, one fragment with EcoRI. The results of the experiments related to the plasmid-elimination and transformation showed the plasmid could have some relationship with Midecamycin-producing in Str. mycaro faciens.
