摘要
文中报道了缺氧条件下培养的肺动脉平滑肌细胞(PASM)的形态特点、生长曲线和蛋白质含量的变化,测定了缺氧时PASM游离钙浓度([Ca^(2+)])和环磷酸腺苷(cAMP)含量的变化.主要结果与结论如下:(1)培养的PASM能在2%O_2缺氧条件下生长,其光镜形态、生长情况与细胞蛋白质含量与2l%O_2常氧条件下相似;(2)向PASM的无钙细胞悬液中分别通入0%O_2、3%O_2和5%O_21min,细胞内[Ca^(2+)]均明显升高(P<0.001),随着通气时间延长,[Ca^(2+)]持续升高至400~513nmol/L;(3)通入N2缺氧1min末PASM的cAMP含量与对照组相比明显升高(P<0.05),5min末高峰(P<0.0l),10min末有下降趋势,但仍高于正常水平(P<0.05).以上结果提示:缺氧(2%O_2)不能导致培养的PASM增生或肥大;培养的PASM对缺氧有较强的耐受力;缺氧可直接影响PASM的第二信使传递系统.
The effects of hypoxia on the growth and the cAMP and cytosolic calcium ions(Ca^(++))contents of the cultured smooth muscle cells of bovine pulmonary artery(PASMCs)were investigated.The findings were as follows:(l)When cultured under an atmosphere containing only 2% oxygen,PASMCs showed similar growth,morphology under optical microscopy and cellular protein content as those under normoxic condition of 21% oxygen.(2)When PASMC suspension in calcium-free medium was exposed to 0%,3% and 5% oxygen for one minute,intracellular ca^(++)level was increased significantly and the increase was more significant when the exposure was prolonged.(3)At the lst minute of anoxia(0%oxygen),cAMP level of PASMCs was significantly increased and the elevation reached the peak at the 5th minute of exposure.At the l0th minute,cAMP level went down but remained higher than the normal control.These findings suggest that PASMCs can relatively withstand hypoxia of 2% oxygen,hypoxia along cannot result in hypertrophy or hyperplasia of PASMCs and hypoxia exerts influence directly on the second messenger system of the cultured PASMCs.
