摘要
目的:观察在高血压不同时期心肌胶原网络的改变,为高血压的治疗及心脏功能的早期保护提供基础资料。方法:用Masson三色法染色、光镜观察和天狼猩红-苦味酸饱和液染色、偏振光显微镜观察两种方法,对压力后负荷增高性高血压大鼠动物模型术后2周、4周、8周心肌胶原进行观察,采用计算机图象分析方法,对高血压不同时期心肌胶原总量和Ⅰ/Ⅲ型胶原比例的改变进行观测。结果:4周时心肌胶原总量较2周时增多,Ⅰ/Ⅲ型胶原量比例无显著改变。8周时,血管周围胶原增生显著,且向周围心肌间质中蔓延,出现了大面积的心肌细胞消失,取而代之为胶原纤维。胶原纤维互相交织,排列杂乱无章。Ⅰ型胶原增多,Ⅰ/Ⅲ型胶原量比例增大。结论:在高血压时,心肌胶原网络重建与血压增速和病程关系密切。血压增高越快,病程越长,胶原网络破坏越严重。
Objective:In order to provide a new way for treating hypertension and the early heart protection, we studied the change of myocardial collagen network during every stage of the spontaneously hypertension. Methods :With the Masson staining with optical microscope and Sirius Red in a supersaturated picric acid solution staining with polarized microscope, myocardial collagen of the rats with pressure-overloaded hypertention about the modality and structure were observed 2 weeks, 4 weeks and 8 weeks respectively after the operation. By using image analysis, we analyzied the change of the total amount of myocardial collagen and the rate of Type Ⅰ collagen to Type Ⅲ. Results:4 weeks After the operation the total of collagen increased more than 2 weeks after the operation, the rate change of type I collagen to type Ⅲ was insignificant. After 8 weeks of operation, the collagen had a big increase around the vascular and spreaded out to myocardium. The collagen increased greatly and myocardial fibrosis was replaced by myocardium in some big areas and the collagen network was irregular. the rate of type Icollagen to type Ⅲ was increased. Conclusion : As the hypertension course lasts,the myocardial collagen network is remodeled. the change is very serious when the time is very long and the blood. pressure rises rapidly.
