摘要
目的:研究大鼠血管内膜损伤后血管重塑的相关因子,包括基质金属蛋白酶(MMP-1,MMP-2),金属蛋白酶组织抑制物(TIMP-2),转化生长因子-1β(TGF-1β)的变化规律。方法:建立大鼠胸主动脉球囊拉伤模型,分为对照组,术后3d、5d、8d、14d、21d和35d组。采用免疫组化方法检测各因子的表达。结果:动脉内膜损伤后3d各因子均明显表达增加,随后下降,TGF-β1至术后8d,MMP-1至术后14d,MMP-2至术后21d降低至与对照组无区别,TIMP-2至术后21d仍持续高于对照。结论:大鼠血管损伤后修复过程中各因子作用时间窗不完全相同,动脉球囊拉伤术后3周内可能是药物干预的最佳时机。
Objectlve:To observe the changes of matrix metalloproteinases (MMP-1,MMP-2),tissue inhibitors of metalloproteinase-2(TIMP-2) and transforming growth factor-β1(TGF-β1)which are remodeling related factors in rat injured arteries.Methods:Stenosis models of thoracic aorta after balloon injury was established in rats. Immunohistochemistry was used to evaluate the expression of above factors at different time. Results:All factors were high expressed obviously on the 3rd day, and started to fall to the level of control, respectively. TGF-β1 persisted for 8 d'ays,MMP-1 for 14 days,MMP-2 for 21 days,while TIMP-2 still kept higher than control till 21st day.Conclusion:All these factors have different time change curve in rat,the first 21 days after injury maybe the optimal period for drug intervention.
