摘要
目的探讨端粒酶活性检测在乳腺癌临床诊治中的意义。方法采用TRAP-assay法检测了人乳腺癌及癌旁正常组织中端粒酶活性。结果细胞株HeLa细胞、K562细胞端粒酶活性均为阳性表达,26例乳腺癌组织端粒酶活性均阳性表达(其中4例呈弱阳性),24例癌旁组织中5例呈弱阳性,19例呈阴性,而lysisbuffer及阴性对照均呈阴性反应。结论端粒酶在人乳腺癌的发生与发展中可能起着重要作用,提示端粒酶活性测定可用于乳腺癌的早期诊断。
Objective To evaluate the role of telomerase activity in development of breast cancer.Methods The telomerase activity in normal tissues、breast cancer and tumor-adjacent tissues were determined using PCR-based TRAP assay.Results Telomerase activity was detectable in HeLa,K562 and breast cancer cells were positive,5 of 24 tumor-adjacent tissue samples were light positive and 19 samples were negative.Conclusion Telomerase may play an important roal in the development of breast cancer.Our results indicate that the detection of telomerase activity may be useful in the diagnosis of breast cancer.
