摘要
作者应用单细胞细胞毒试验及形态学观察,研究多抗甲素(PAA)对人体自然杀伤(NK)细胞活性的作用机理。结果表明:PAA能增强食道癌患者淋巴细胞(PBL)明显低下的结合率(B%)和杀伤率(K%),也可增强正常人PBL的K%,但对正常人PBL的B%无影响。形态学观察发现:与靶细胞结合的PBL除大颗粒PBL外,尚有普通的非颗粒PBL,结合的靶细胞损伤首先表现为线粒体肿胀、空泡变,然后核固缩,核膜损伤,细胞严重空泡变性等,偶见靶细胞与自然杀伤细胞结合部质膜破损的现象。经PAA预处理的靶效细胞结合的形态改变无明显差异,但食道癌组的杀伤作用出现较早,且明显增强。
It has been shown that the novel im-munopotentiator Polyactin A(PAA)developed inChina can augment NK cell activity. In order to fur-ther understand the mechanism of PAA effect on NKactivities ,we measured the binding and lytic capacitiesof peripheral blood lymphocytes( PBL)isolated from normal subjects and patients with esophageal carcino-ma against K_(562) target cells by using a single cell cyto-toxicity assay. Morphologically the ultrastructures ofNK cells of normal subjects and the patients on PAAwere also studied. The results showed that:(1)Prein-cubation PBL of normal subjects on PAA at 100 μg/mlfor 4 hours could increase the rate of NK lysis with noeffect on the binding rate.The binding capacities ofPBL of patients with esophageal carcinoma were lowerthan those of normal subjects. Pretreatment PBL ofpatients with esophageal carcinoma could significantlyincrease not only the rate of NK lysis, but also thebinding rate.(2) Study on the processes of conjuga-tion and cytolysis by using light microscopic morpholo-gy,scanning electron microscopy (SEM ) and transmis-sion electron microscopy(TEM ) found that effectorcells include not only large granular lymphocytes(LGL) ,but also non-LGL bound to target cells.Oneeffector cell could simultaneously make contacts withmore than one target cell. It was found that PBL/K_(562)contacts could be morphologically divided into twotypes in the SEM and TEM.Either effector cell pro-trusions were pushed deep into pouches and lacunae ofthe target cell surface or a wide area of intimate cell-to-cell contact was formed. Effector/target cell con-tact is a prerequisite for NK lytic activity. After theformation of contact,alterations of the target cell fol-lowed.During the disintegration stage,internal vac-uolization and svvelling of cytoplasmic organelles,par-ticularly welling of mitochondria occurred. Eventual-ly , karyopyknosis,karyothecalysis,and honeycomboccurred in the target cell.Sometimes local cy-topathogenic membrane or cytoplasmic changes wereobserved in the contact areas. In conclusion,the lysisof NK targets may be due to not only the direct con-tact between effector and target cells,but also NKlytic factors.
