~3H-GABA和~3H-Glu在海马的摄取及其衰老性变化 被引量:2

DISTRIBUTION AND AGING CHANGES OF THE UPTAKE SITES OF ~3H-GABA AND ~3H-GLUTAMATE IN HIPPOCAMPUS

摘要 用体外摄取和放射自显影方法研究~3H—GABA和~3H—Glu摄取位点在大鼠海马的分布和摄取量的衰老性变化。GABA摄取位点主要位于锥体细胞和颗粒细胞的周围以及放射层,与篮细胞突起的分布相一致。Glu摄取位点主要定位在海马穿通通路,苔藓纤维和Schaffer侧枝的投射区。衰老时海马对GABA摄取增加,p<0.01;对Glu摄取减少,p<0.01。GABA摄取增多可能和胶质细胞反应有关,而Glu摄取减少对于解释衰老性海马神经元损害和电生理改变的机制提供了新的线索。 Gamma-aminobutyrate (GABA) and Glutamate (Glu) both are important transmitters in hippocampus. They seem to assume inhibitory and excitatory action respectively in hippocampal loop. Glu is closely related to the development and the sustenance of frequency potentiation and long term potentiation (LTP) in hippocampus. During senes cence hippocampal frequency potentiation and LTP are affacted but it is rot clear whether GABA and Glu in hippocampus change. Distribution and aging changes of the uptake sites of GABA and Glu were studied by the autoradiographic approach in hippocampus of adult and aged Wistar rats. Distribution of the GABA uptake sites consisted with the (?)ocation of basket cells and their axon terminals. Glu uptake sites were mainly located in the target areas of perforant path, mossy fibers and Schaffer collaterals. GABA uptake sites (Silver grains/500μm^2) in CA_1 and CA_3 areas of hippocampus were more in aged group (30 months) than in adult group (12 months), P<0.01. In contrast with GABA, Glu uptake sites were less in aged group than in adult group, P<0.01. The results suggest that the uptake changes of GABA and Glu might be caused by glia cell reactivity and neuron loss respectively, and that Glu uptake decrease contributed to neuronal damage and electrophysiologic variety in hippocampus during aging.
