摘要
本实验分别将日本血吸虫(大陆株)雌、雄、合抱虫体可溶性抗原制剂,佐以卡介苗,一次性皮内免疫小鼠。结果,免疫后的小鼠对血吸虫的攻击产生相当的保护作用(保护率分别为42.3%、42.6%和44.5%)。经SDS-PAGE和Western Blot对虫体可溶性抗原进行分析,免疫血清为1:300时,仅有一条蛋白带出现强而明显的反应,其反应蛋白带的分子量为94/90kD。
Soluble adult worm antigens (SWAP) of Schistosoma japonicum were used to im- munize mice against challenge infection. Adults of female, male and pair of the worms were homogenized, iced and thawed, fractured under supersonic treatment and then centrifuged, respectively; The supernatants containing the soluble antigens (SWAP_f,SWAP_m,SWAP_p) and BCG were used to immunize mice weighed 18~ 20g intradermally at chest with total 0. 1ml solution containing 1mg SWAP and 1 mg BCG as a adjuvant for each mouse. 4 weeks after immunization each mouse was challenged with 40 cercariae and 6 weeks later the mice were killed and the number of burden worms was recorded. The results indicated that Use of SWAP in conjunction with BDG resulted in a strong protec- tive effect against challenge of parasites. The reduction rates in worm burdern in the three experi- menytal groups were 42. 3%, 42. 6% and 44. 5%respectively. The result of ELISA demonstrated strong reactions of the immune sera to schistosoma anti- gens. In Western Blot analyses of SWAP, 4 positive bands appeared when the immnue sera were 1:100 dilution,of which 94/90KD demonstrated the strongest reaction,this band contained almost 60% of the total reacted protein. When immune sera of 1:200 or 1:300 were chosen, only one sin- gle 94/90KD band appeared. Therefore,the 94/90KD protein in SWAP antigens was likely to play a major role in the development of protective immunity against challenge infection.
