摘要
目的 探讨线粒体钙浓度 (MCC)和能量代谢变化之间的相互关系以及细胞内钙浓度升高的来源。方法 建立不同Ca2 + 浓度 (3mmol/L、 1. 5mmol/L)及不同培养时间 (2h、 4h)条件下大鼠离体骨骼肌被动牵拉培养的模型 ,测定线粒体钙浓度 (MCC)、ATP酶活性和乳酸 (LA)含量。结果 MCC在各实验中实验组与对照组相比均有显著性增加 (P <0 . 0 5 ) ;LA含量在 3mmol/LCa2 + 培养液中培养 2h条件下有显著增加 (P <0 . 0 5 ) ,在其他实验中均无显著性变化 ,Na+ - K+ ATPase、Ca2 + - ATPase亦无显著性变化。结论 MCC在牵拉负荷条件下有显著增加 ;胞内钙平衡紊乱可能发生在能量代谢障碍之前 ,并成为肌肉损伤的初始因素。
Objective The purpose of the study is to explore the correlation between mitochondrial calcium concentration(MCC)and cellular energy metabolism.Method 40 rats were divided into 5 groups according to the calcium concentrations and the incubation times,the isolated soleuses in vitro incubated in 3 mmol/L Ca 2+ buffer for 2 hours(group Ⅰ) and 4 hours(group Ⅱ),in 1.5 mmol/L Ca 2+ buffer for 2 hours(group Ⅲ),and 4 hours(group Ⅳ);the soleuses incubated in Ca 2+ -free buffer for 2 hours were the controls,all muscles were under stretch conditions.After incubation,measure the levels of mitochondrial calcium(MCC),lactic acid(LA)and the activities of ATPase in the muscles.Result The results showed that the MCC level was elevated significantly in all experimental groups(P<0.05 compared with control group),LA level only showed some increase in group Ⅰ(P<0.05),while Na +-K +-ATPase and Ca 2+ -ATPase activities had no any significant change in all experimental groups.Conclusion Stretch load could elevate the MCC of skeletal muscles,and the failure of intracellular calcium homeostasis might occur before the malfunction of energy metabolism,that should be the initiating factor of muscle injury according to this study.
