摘要
目的 探讨建立合适的实验性糖尿病性聋动物模型的方法。方法 应用Wistar大鼠经阴茎静脉注射不同剂量链脲佐菌素 (streptozotocin ,STZ)诱导建立实验性糖尿病性聋动物模型 ,观察实验动物血糖、尿糖、体重及听性脑干反应变化情况。结果 STZ 4 0mg/kg组动物糖尿病成模率仅为 2 2 % ,且成模动物饲养 3个月后仍无听力损害情况出现。而STZ 5 5mg/kg组及 6 5mg/kg组动物糖尿病成模率分别可达 94 %及 95 % ,且糖尿病成模后 2月起即有听力损害情况。至成模后 3个月 ,听力损害情况更进一步加重。STZ 6 5mg/kg组动物死亡率极高。结论 实验显示我们建立的实验性链脲佐菌素糖尿病性聋动物模型是成功的 ,5 5mg/kg的STZ剂量为诱导实验性糖尿病性聋大鼠模型。
Objective To investigate the method of establishing an ideal experimental hearing impaired diabetes animal model.Methods Wistar rats were injected intravenously with different dosage of streptozotocin to induce experimental animal model. Results were evaluated by measurement of blood glucose, urine glucose, body weight and auditory brainstem response.Results STZ 40 mg/kg could only induce 22% diabetic rat model and failed to have hearing impairment after 3 months of diabetes modeling. STZ 55 mg/kg and 65 mg/kg could respectively evoke 94% and 95% diabetic rat model with significant hearing impairment, but in STZ 65 mg/kg group animals had much higher mortality rate.Conclusion The experimental hearing impaired diabetes animal model was successfully established, STZ 55 mg/kg was the proper dosage for evoking the animal model and using for long term research.
