摘要
目的 研究多发性硬化 ( MS)患者外周血 ( PB)中自然杀伤 ( NK)细胞的表达。方法 采用流式细胞仪技术 ,检测 1 5例 MS患者以白细胞介素 -1 2 ( IL-1 2 )刺激 NK细胞增殖前后 PB NK细胞的百分率。结果 MS患者PB中 NK细胞百分率明显低于对照组 ( P <0 .0 5 ) ;以不同浓度的 IL-1 2刺激 NK细胞增殖后 NK细胞百分率呈剂量依赖性增加 ,于 5 d时达到峰值 ,但较刺激前明显减少 ( P <0 .0 5 )。结论 MS患者 PB中 NK细胞的表达明显减少 ,以 CD1 6+ 和 CD5 6+ 细胞减少为主 ;IL-1 2在 1 0 2 U/m L数量级时可对 NK细胞增殖活性产生最佳激活作用。
Objective The expression of natural killer(NK) cells were studied in peripheral blood(PB) of multiple sclerosis(MS) patients.Methods The percentage of NK cells in PB stimulated by interferon-12(IL-12) of different concentrations were measured in 15 MS patients by using fluorescence-activated cell sorter(FACS).Results MS patients had lower percentage in PB than control groups (P<0.05); NK cells showed dose-dependence when stimulated with different concentrations of IL-12, the peaks were observed on the 5th day, and the percentage was significantly decreased(P<0.05).Conclusions NK cells expression decreased markedly in MS patients, CD16+ and CD56+ NK cell was the main influenced subset which decreased more significantly. The best order of magnitude of IL-12 to promote NK cells proliferation was 102 U/mL.
